Can we set up an auto-reply with the instructor's course email account? If so, how?

I'm not 1000% across the context for the auto-reply, however the Intelligent Agent tool can be used to send email to learners based on their usage behaviour in the course.

When I am on holiday (Christmas or Spring break), I would like my email to automatically reply to a student who has emailed me saying, "…I will be back on …"
